Lion's mane is a shaggy white mushroom that grows on hardwoods across the Northern Hemisphere, eaten in East Asia (it cooks like crab) and used in Chinese medicine for digestive complaints. Its modern fame comes from laboratory work: compounds in the fruiting body (hericenones) and mycelium (erinacines) stimulate synthesis of nerve growth factor (NGF), a protein essential to neuron survival and growth. Evidence by use

Cognition — the load-bearing trial. A 2009 double-blind Japanese study randomized 30 adults aged 50–80 with mild cognitive impairment to 3 g/day of fruiting-body powder or placebo for 16 weeks: cognitive scores rose progressively versus placebo, then declined toward baseline four weeks after stopping. Encouraging design, honest result — and sixteen years later, still essentially unreplicated at scale. A handful of tiny subsequent studies in healthy adults (single doses or a few weeks) show scattered, inconsistent improvements on isolated tasks.

Mood. A 4-week trial in 30 menopausal women found reduced anxiety and irritability scores on a symptom questionnaire versus placebo. Small, short, one population.

Nerve repair, neuropathy, MS, depression mechanisms — animal and laboratory territory so far. For neuropathy in particular, ingredients with actual human trials exist (alpha-lipoic acid); lion's mane is not among them.

As food, long and reassuring history. As a supplement in trials: well tolerated, with mild digestive upset the main report. Cautions:

  • Allergy — case reports of rash and breathing difficulty; anyone with mushroom allergy should avoid it.
  • Blood thinners — laboratory antiplatelet activity; no human bleeding cases established, but mention it alongside warfarin, DOACs or aspirin, and stop before surgery on general principle.
  • Diabetes medication — animal glucose-lowering data; monitor if combining.
  • Pregnancy and breastfeeding — no data; avoid.
